Intended Use

The HDL-EX "SEIKEN" kit is for the quantitative determination of smaller and denser HDL (HDL3) cholesterol (-C) in human serum or plasma

Clinical Significance

HDL comprises of several subclass particles, which differ in their sizes, densities, and components. These HDL subclasses are considered to play different roles in progression and regression of arteriosclerosis. The HDL3-EX “SEIKEN” kit enables a fully automated assay for quantification of HDL3-C. The test is completed within 10 minutes.

Principle

The assay consists of two reaction steps. In the first step, chylomicrons, VLDL, IDL, and LDL are decomposed by a surfactant and sphingomyelinase (SPC). The cholesterol released from these lipoprotein particles is then degraded finally to water and oxygen through the hydrolysis by the cholesterol esterase (CHE), oxidization by the cholesterol oxidase (CO), and subsequent reaction of catalase against intermediately produced hydrogen peroxides during the reaction.

In the second step, a specific surfactant selectively reacts with HDL3 among the remaining lipoproteins, releasing cholesterol specifically from HDL3. The catalase in the reaction mixture is inhibited by sodium azide in Reagent-2. The hydrogen peroxides produced from the reaction of the released cholesterol with the CHE and CO, develop a purple-red colour with the coupler in the presence of peroxidase (POD).
