Intended Use

For the quantitative in vitro determination of Alkaline Phosphatase in serum or plasma. This product utilizes the AMP method and is suitable for use on the RX Series instruments which includes the Rx Daytona and the Rx Imola.

Clinical Significance

Serum Alkaline Phosphatase (ALP) measurements are of diagnostic use in the investigation and treatment of hepatobiliary disease such as hepatitis. In liver disease damaged liver cells release high levels of ALP into the blood. ALP levels are usually much lower than both AST and ALT however when bile ducts become blocked due to gallstones etc ALP levels are much higher than both AST and ALT.

ALP levels are also elevated in any condition that affects bone growth and in bone disease associated with increased osteoblastic activity. ALP can be used to help detect certain types of cancer that may have spread to the bone and bone conditions such as Paget’s disease where the boned become deformed. Also useful in monitoring cancer treatment.

ALP levels are also useful in the diagnosis of parathyroid and intestinal diseases.


This is an optimized standard method according to the recommendations of AACC and IFCC.

The substrate p­nitrophenyl phosphate is hydrolyzed by Alkaline Phosphatase from the sample, in the presence of Magnesium ions, to form p­nitrophenol which is yellow in color and can be read at 405 nm. The intensity of color produced is proportional to the Alkaline Phosphatase activity in the sample.
